BioRad's Bacterial Transformation (pGLO) Lab

Participants transformed a strain of E.coli using calcium chloride and heat shock which made the bacteria susceptible to accepting a foreign plasmid. The genetically engineered plasmid consisted of the glowing green jellyfish gene, an antibiotic resistant gene, and a regulator gene for arabinose (sugar). If the transformation was successful, the LB/amp/ara plate will contain bacteria that will glow in the presence of UV light. Bacteria growing on an ampicillin plate took up the plasmid but will not glow if arabinose is not present in the agar plate. GREAT lab for demonstrating the effects of a regulator gene.

March 23: Teachers and parents extracted DNA from their favorite snacks to test for the "Bt" using primers for the CaMV 35S promoter and NOS terminator.

Notice the difference a filter can make when looking at Gel 2. Lane 1 & 2 have faint bands (Blaine's Sun Chip).

BioRad GMO Investigator Kit (#166-2500)

 Feb. 26 - March 1:   Biomed students tested favorite snacks for being genetically modified. Using primers for the Cauliflower Mosic Virus 35S promoter (203 bp) and Chloroplast component gene (455 bp), students were able to successfully purify plant DNA and determine the presence of the "Bt" gene.

Holmes Community College - Ridgeland hosts Wolbachia Rodeo

February 1, 2013 - The 3rd Mississippi High School Wolbachia Rodeo was hosted by Holmes Community College with coordination between Joe David White (Holmes CC) and Betsy Sullivan (Madison Central High School). Twenty-two students from 3 different high schools participated (Germantown, Madison Central, and St. Andrew's).
